Estrone is a natural follicular hormone that regulates the normal development of the female reproductive system and is produced in the ovaries with the onset of puberty before the onset of menopause.
The action is mediated through estrogen-sensitive receptors of the genitals, mammary glands, hypothalamus and pituitary gland. It has a specific estrogenic effect - it stimulates the proliferation of the endometrium (in the fallopian tubes, cervix, vagina, external genitalia, excretory ducts of the mammary glands), the development of the uterus and secondary female sexual characteristics when they are underdeveloped, eliminates or reduces the severity of disorders that occur in a woman’s body in connection with insufficient function of the gonads during menopause or after gynecological operations. Together with progesterone, it participates in the regulation of the menstrual cycle and the function of childbirth.
In small doses it activates the secretion of follicle-stimulating hormone, in large doses it inhibits it, and also retains water and sodium ions in the body, inhibiting erythropoiesis. Pharmacokinetics

Well absorbed. High plasma levels persist for 4-5 days. Metabolized mainly in the liver (partially in the lungs, adipose tissue), excreted in bile, reabsorbed in the intestine and again enters the liver through the portal system. Contraindications

Directions for use and doses

Prescribed intramuscularly. The dose is set individually.
Primary amenorrhea with underdevelopment of the genital organs and secondary sexual characteristics. Prescribe 1-2 ml daily or every other day for 1-2 months or more (until noticeable enlargement of the uterus). Then progesterone is prescribed (intramuscular, 5 mg daily for 6-8 days). If necessary, courses of hormone therapy are repeated.
Secondary amenorrhea. Prescribe 1 ml daily for 15-16 days, followed by progesterone for 6-8 days. If there is no lasting effect, the course of treatment is repeated.
Hypo- and oligomenorrhea, algomenorrhea, infertility caused by ovarian hypofunction or underdevelopment of the uterus. After the end of menstruation, 0.5-1 ml is prescribed daily for 15-16 days, then, if indicated, progesterone is prescribed for 6-8 days. The course of treatment can be repeated at the same time after the end of menstruation. Functional disorders caused by the onset of menopause and oophorectomy (depression, angioedema, etc.). Prescribe 0.5-1 ml daily or every 1-2 days in courses of 10-15 injections. The course of treatment is repeated if symptoms recur. Weakness of labor and post-term pregnancy. Prescribe 4-5 ml 2-3 hours before using birth stimulants.

Side effect

Nausea, vomiting, headache, dizziness, allergic reactions, eyelid swelling, erythropenia. With long-term use, uterine bleeding, ovarian sclerosis, disturbances in the metabolism of sodium, calcium and water, and congestive jaundice are possible.

Overdose

Symptoms: increased headache, nausea. With prolonged use, uterine bleeding is possible. Treatment: symptomatic.

Interaction with other drugs

Enhances the effect of hypocholesterolemic, antiarrhythmic, antihypertensive drugs, diuretics, anticoagulants, tricyclic antidepressants, glucocorticoids, thyroid hormones. The effect of estrone is enhanced by folic acid and thyroid medications, and weakened by antiestrogenic drugs (clomiphene, etc.) used to treat infertility, phenobarbital, rifampicin and other monooxygenase inducers. The concentration of the drug in the blood is reduced by ampicillin, chloramphenicol, tetracycline, neomycin, sulfonamides; increases - ascorbic acid.

The drug is prescribed with caution for arterial hypertension, previous thromboembolic diseases, epilepsy, depression (including a history), gallbladder diseases, otosclerosis.

Interaction: Ampicillin, chloramphenicol, tetracycline, neomycin, sulfonamides reduce the concentration in the blood (reduce enterohepatic circulation), ascorbic acid increases. Enhances the effect of tricyclic antidepressants (slows down inactivation in the liver), glucocorticoids, and thyroid hormones. Directions for use and dosage: IM. For replacement therapy - 0.1-1 mg per week, once or in several doses, from the 1st to the 25th day of each month, with a 7-10-day break, 2-3 courses.
